Victoria Hand first appeared in the Marvel universe as an openly lesbian character strongly opposed to the way S.H.I.E.L.D., for whom she was employed as an accountant, was run under Nick Fury. After voicing her opinion in a letter, she was shipped out to Oregon to a remote S.H.I.E.L.D. location. From there her career in S.H.I.E.L.D. took a pretty fortuitous turn when Norman Osborne, who almost certainly won't be appearing on Agents of S.H.I.E.L.D., was named head of the organization which he renamed H.A.M.M.E.R. Osborne appointed Hand Deputy Director of H.A.M.M.E.R., a role in which she found herself supporting villains and hindering heroes in support of Osborne who she felt was doing the right thing.Without going too far into detail that may spoil her role on the show, Hand
eventually redeemed herself before her death.
While her role is undefined on the show, it's probably not a long shot to assume that she may be a part of changing of the guard at S.H.I.E.L.D. that leads to Alexander Pierce eventually taking charge of the organization. Clearly the S.H.I.E.L.D. we see in the Cap trailer is not a stable organization and Fury is on the run. It's possible that we see Hand play a part in that during in upcoming episodes of the show.
